水曲柳丙二烯氧化物合成酶基因FmAOS序列与表达模式分析

摘 要:

茉莉酸(jasmonic acid, JA)作为一种重要的信号分子参与了植物的生长发育过程, JA生物合成途径中的关键酶为丙二烯氧化物合成酶(allene oxide synthase, AOS)。本研究克隆了水曲柳(Fraxinus mandschurica) AOS基因, 命名为FmAOS。生物信息学分析表明该基因全长1 605 bp, 具有完整的开放阅读框, 编码534个氨基酸。FmAOS为两性不稳定性蛋白, 不存在信号肽, 具有跨膜能力, 亚细胞定位预测其存在于叶绿体中。三维建模分析表明该蛋白属于混和型。FmAOS表达模式分析表明, 其响应了寒冷、NaCl、干旱等非生物胁迫及脱落酸(ABA)、赤霉素(GA3)等植物激素信号, 但表达模式各不相同, 说明FmAOS参与了植物的生长发育和逆境胁迫的平衡。本文为FmAOS功能的深入研究提供参考。

Sequence and expression analyses of allene oxide synthase gene FmAOS in Fraxinus mandschurica

Abstract:

Jasmonic acid (JA) is an important signal molecule participating in plant growth and development. Allene oxide synthase (AOS) is a key enzyme in JA biosynthesis. We cloned AOS gene in ash tree (Fraxinus mandschurica), and named it as FmAOS. Bioinformatic analysis showed that FmAOS is 1 605 bp, containing a complete open reading frame, encoding 534 amino acids. FmAOS protein is amphoteric and unstable, has no signal peptide but transmembrane ability, and exists in chloroplast by subcellular localization prediction. According three dimension modeling analysis, FmAOS is belong to mixed protein. Expression analysis showed FmAOS responds to abiotic stress such as cold, NaCl, drought and plant hormone signals like ABA and GA3, but the expression patterns are different, which means FmAOS is involved in the balance of growth/development and stress response. This paper can lay a foundation for the study of FmAOS gene function.
